Our SyGreen qPCR master mix utilizes the latest advances in polymerase technology and buffer chemistry to provide outstanding results on any DNA template including genomic, cDNA and viral sequences with the greatest efficiency. This all-in-one master mix requires little to no optimization. The 2x master mix uses an antibody-mediated hot start system that allows for a quick 2 minute activation step. This proprietary small molecular inhibitor technology prevents formation of primer dimers and non-specific products leading to improved reaction sensitivity and specificity.
The qPCRBIO SyGreen Mix utilizes a non-PCR inhibiting intercalating dye. The blue mix contains a non-reactive blue dye that allows for easy visualization during pipetting. Extremely low copy number targets can be detected specifically with the highest efficiency. The same developments that improve the sensitivity and consistency of qPCRBIO SyGreen Mix in standard cycling conditions also allow for industry leading performance in fast and ultra fast cycling conditions on any real-time PCR cycler. SYBR and SyGreen are names that refer to the dye used in the master mix. The SyGreen 2 dye used in our SyGreen mix belongs to the same family of dyes that the SYBR green dye belongs to (asymmetric cyanine dyes). The SyGreen 2 dye is an improved, new generation dye. Both dyes intercalates the DNA, however SyGreen 2 supports pCR amplification over a wide range of dye concentrations and the dye does not inhibit the PCR reactions, unlike SYBR green. This plays a role in producing robust and consistent DNA melting curves both with prokaryote and eukaryote templates, as it shows no preference in nucleotide composition.
Yes, products can be sequenced or digested with restriction endonucleases using standard protocols. Products produced are 3'-d(A)-tailed and can be used for TA cloning. They also may be blunt-ended or digested with restriciton enzymes prior to cloning. We recommend purifying your PCR products using a PCR clean-up kit such as the Zymo DNA Clean & Concentrator kit that we offer.
Rox is a passive reference dye that does not take part in the PCR reaction. It is used to normalize non-PCR reated fluctuations in fluorescence. Please see the Instrument Compatability Guide to determine which Rox level is right for your qPCR machine.
An early Ct could indicate one of two things. The first could be the result of our product performing very well relative to the competitors product. We have designed our products to be faster than our competitors, and we have found that we are significantly faster than many. The second could be that caused by non-specific products. This can be verified by running a melt analysis at the end of the qPCR run. If your results show one peak, it is likely the correct product was amplified. You may observe the position of the peak being different with our mix relative to our competitors mix but is this normal because the salt concentration and pH affect the melting temperature. If your results show more than one peak, then the reaction is not specific. PCR products can also be run on an agarose gel to verify if there is only one band, and if it is the same size as the amplicon obtained with the competitors product. To address the issue of a non-specific product, the thermal cycling will need to be adjusted.
